Indeed it is possible. There were several cards made which had two floppy ports, including the famous Compaticard.
I still have such an arrangement in one of my DOS boxes, though I'm not using it at present. The expansion card itself had special BIOS on it, which allowed DOS to recognize the additional floppies (MS-DOS, by itself, would not do this).
I don't believe that DOS will recognize two separate controllers. Other OS's might.
